GOLFa Mediates Dopamine D1 Receptor Signaling

چکیده

It is generally assumed that the coupling of dopamine D1 receptors to adenylyl cyclase is mediated by the stimulatory GTP-binding protein Gs. However, the striatum contains little Gsa subunit, whereas it expresses high levels of Golfa, a close relative of Gsa that is also expressed in olfactory receptor neurons. We used Golfa knockout mice to examine the functional coupling of D1 receptors. We found that these mice showed no hyperlocomotor response to either the D1 agonist SKF-81297 or the psychostimulant cocaine. Moreover, Golfa knockout mice did not display cocaine-induced c-fos expression in the striatum. Finally, in the absence of Golfa, striatal D1 receptors have a decreased affinity for dopamine. Thus coupling to Golfa appears to mediate D1 signaling in the striatum.

D1-like dopaminergic activation of phosphoinositide hydrolysis is independent of D1A dopamine receptors: evidence from D1A knockout mice.

Accumulated evidence suggests that dopamine and dopamine D1 agonists can activate phospholipase C in both brain and peripheral tissue. The receptor that mediates the hydrolysis of phosphoinositides has not been identified. The cloned dopamine D1A receptor that is generally thought to be linked to adenylyl cyclase, has also been proposed to couple to phospholipase C. However, a number of studies...

Trk B signaling in dopamine 1 receptor neurons regulates food intake and body weight

OBJECTIVE Loss of BDNF-TrkB signaling results in obesity in both humans and mice; however, the neural circuit that mediates this effect is unknown. The role of TrkB signaling in dopamine-1 receptor expressing neurons in body weight regulation was tested. METHODS Mice with a floxed allele of the TrkB gene were paired with mice expressing Cre-recombinase under control of the D1 promoter to cond...

MicroRNA 142-3p Mediates Post-Transcriptional Regulation of D1 Dopamine Receptor Expression

The D1 dopamine receptor subtype is expressed in the brain, kidney and lymphocytes. D1 receptor function has been extensively studied and the receptor has been shown to modulate a wide range of physiological functions and behaviors. The expression of D1 receptor is known to change during development, disease states and chronic treatment; however, the molecular mechanisms that mediate the change...

Study of Dopamine Receptor Gene Polymorphisms and Their Association with Growth and Egg Production Traits in West Azerbaijan Native Chicken

The objective of this study was to search for single nucleotide polymorphism (SNP)-type polymorphisms in the dopamine D1 receptor in West Azerbaijani native chicken and look for their association with egg production and body weight traits of chickens by polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP). For this purpose 180 blood samples were taken from nativ...

Dopamine receptor 1 neurons in the dorsal striatum regulate food anticipatory circadian activity rhythms in mice

Daily rhythms of food anticipatory activity (FAA) are regulated independently of the suprachiasmatic nucleus, which mediates entrainment of rhythms to light, but the neural circuits that establish FAA remain elusive. In this study, we show that mice lacking the dopamine D1 receptor (D1R KO mice) manifest greatly reduced FAA, whereas mice lacking the dopamine D2 receptor have normal FAA. To dete...
